Sterile RPMI-1640 was developed for the cultivation of normal and neoplastic human leukocytes and is based on modified amounts of amino acids and vitamins. RPMI-1640 Medium is very well suited for culturing of a wide range of mammalian suspension cell lines and primary lymphocytes, all leucocytes, B- and T-lymphocytes, as well as myeloma- and hybridoma cells. Generally, FBS is added to RPMIl media. However, it has been shown that RPMI media also support several cell lines in the absence of serum. ROTI ® Cell RPMI-1640 CELLPURE ® ready-to-use, sterile, with stable glutamine This modification of RPMI contains stable glutamine, but no pyruvate. Stable glutamine is present as the dipeptide L-alanyl-L-glutamine. Due to its reinforced structure, this form of glutamine is not temperature sensitive, unlike normal L-glutamine, and therefore does not spontaneously decompose to toxic ammonia. Cells are able to break down this dipeptide bond and release L-glutamine as needed. Thus, the low ammonia concentration improves cell viability and growth. In addition, the concentration of L-glutamine is kept constant. Stable gluatmine has already been added to some cell culture media, but can also be purchased as a separate additive solution, the ROTI ® Cell Glutamine Solution.
